Main purpose of Lab:
This lab is mainly emphasizes on how we collect water sample from three different locations such as paddle boat, fish dock and finally from Gees area. The sample analyzed in the lab measure the turbidity and also measures the bacterial colonies (Escherichia Coli and Coliform bacteria).
E. coli (Escherichia coli) are an adherent of the coliform group of bacteria and are present simply in the warm-blooded animal’s intestines, counting humans. When present in drinking water, E. coli specifies the water has been polluted with animal or human wastes (feces). 6. Take 10 ml sample water from site and after that filled up to 100 with sterile water and add into the filter membrane.
7. For total coliform bacteria placed sample in table incubator for 350C for round about 24 hrs.
8. For fecal bacteria placed sample in water bath for 44.50 C for round about 24 hrs.
9. For TOT count red and blue points in the dish.
10. For fecal bacteria count dark blue points.
11. Repeat the same experiment for almost 7 more times for every three locations and measure temperature, turbidity with turbidity meter and counts the number of bacterial colonies.

Measuring turbidity in ecological uses, such as lakes, oceans and rivers, a Secchi disk can be used. This is a white and black disk that is pull down into the water till it can no extended be seen.  The depth is called Secchi depth the correlating number is noted as a quantity of the clearness in the water. The benefit in using this method in open waters is the capability to measure turbidity at numerous depths of despair wherever numerous turbidity layers are extant. This instrument is also cool to use and comparatively low-cost. As we can see from the above results maximum turbidity present in the paddle boat, then fish dock and least turbid water was present in the gees water site location.
